Azinomycins A and B, new antitumor antibiotics. I. Producing organism, fermentation, isolation, and characterization.

چکیده

A strain of Streptomyces griseofuscus S42227 (FERM P-8443) was found to produce new antitumor antibiotics, called azinomycins A and B. The molecular formulas of azinomycins A and B were determined as C30H33N3O10 and C31H33N3O11, respectively. They were active against Gram-positive bacteria, Gram-negative bacteria and L5178Y cells in tissue culture.

A new antibiotic, cyanocycline A, was isolated from the fermentation broth of Streptomyces flavogriseus strain No. 49, a soil isolate. The molecular formula of cyanocycline A was determined to be C22H26N4O5. The antibiotic has a cyano group and a N-heterocyclic quinone moiety in its structure. Cyanocycline A was found to have broad spectrum antimicrobial and antitumor activity.

A new antitumor antibiotic SF2575 has been isolated from a culture filtrate of Streptomyces sp. SF2575. The molecular formula was determined to be C40H43NO15 by elemental analysis, mass and 13C NMR spectral analyses. The spectral data revealed SF2575 to be a new tetracycline antibiotic. It was active against Gram-positive bacteria and exhibited antitumor activity against P388 leukemia in mice.

The new antibiotics 4181-A and B were isolated from the fermentation broth of Streptomyces griseus, a soil isolate. Their molecular formulae were determined as C29H21NO9 and C28H19NO9, respectively. The UV, IR and NMR spectra suggest that they possess a quinone moiety in their structures. They were found to have antibacterial, antifungal and antitumor activity.
